Mitragynine (MG)

The primary alkaloid in Mitragyna speciosa, responsible for most of kratom’s mild stimulant and relaxant effects at typical doses.

Why it matters

Mitragynine accounts for roughly 60–70% of the alkaloid content in a dried kratom leaf. It is a partial agonist at the mu-opioid receptor and also acts on adrenergic, serotonergic, and dopaminergic systems — a polypharmacology that explains why low doses tend to feel stimulating and higher doses sedating. A January 2025 study published in Nature Chemistry by University of Florida researchers characterised mitragynine as a “biased agonist” at the mu-opioid receptor: it activates pain-relieving pathways while showing significantly weaker recruitment of beta-arrestin, the signalling pathway linked to respiratory depression in classical opioids.

In practice

In a kratom Certificate of Analysis, mitragynine concentration is usually reported as a percentage of total dry weight (typically 1–2% in commercial powder). Vendors who batch-test will publish that percentage on every batch label. Practically, that number lets the consumer compare relative potency across products and across vendors. It is not a substitute for dose-finding — variability in absorption and metabolism means two people taking the same milligrams of mitragynine can experience different effects.

Common confusion

Mitragynine is sometimes confused with its more potent metabolite 7-hydroxymitragynine (7-OH). The two are pharmacologically distinct: mitragynine is the parent compound, abundant in the leaf and milder per milligram; 7-OH is present in trace amounts in the raw leaf but is also generated when the body metabolises mitragynine. Concentrated 7-OH products marketed as “extracts” are not the same product class as leaf powder.
