Kratom tolerance

The pharmacological adaptation that reduces the effect of a given kratom dose with regular use — driven primarily by mu-opioid receptor downregulation.

Why it matters

With repeated exposure, mu-opioid receptors downregulate and intracellular signalling pathways desensitise. The user’s subjective response to a given dose decreases. Tolerance can develop within days of daily use and plateau within weeks, depending on dose, frequency, and the specific alkaloid profile being consumed. 7-OH-heavy products produce tolerance more aggressively than leaf-only products.

In practice

The two practical management strategies are dose holidays (full breaks long enough for receptor density to recover, typically days to weeks) and rotation (alternating between leaf strains with meaningfully different alkaloid profiles). Tolerance breaks are more effective than rotation; rotation alone tends to slow tolerance development rather than reverse it.

Common confusion

Tolerance is sometimes conflated with addiction. They are related but distinct: tolerance is a pharmacological adaptation that can occur without behavioural compulsion; addiction is a behavioural syndrome that can occur with or without dramatic tolerance.
